My question concerns exposure times and fluorescence values.
I am imaging brain slices under an Olympus BX50 fluorescence microscope with a FITC cube & mercury burner. I have brain slices that have been covered in Zinpyr-1 (a fluorescent zinc probe) that binds to "unbound" or "free" zinc.
I plan on analyzing these images that I have taken with ImageJ and had a question regarding whether or not comparisons between those pictures taken at 1/11sec exposure times are comparable with 1/5.0sec exposure times. Should I only use images in comparison that have the exact same exposure time used to capture the image? I'm keeping a lot of things consistent (green levels, objective lenses) but have noticed that changing from 1/11 to 1/5 leads to a brighter image but whether or not that affects the fluorescence values on ImageJ is still an issue.
I'm new to the area of fluorescence but plan on taking images and gathering AFUs (artificial fluorescence units) from the hippocampus.
