Injection of CD4-specific antibodies (i.p. or i.v.) has been widely employed as a method of depleting CD4+ T cells. However, plasmacytoid dendritic cells are also known to have CD4 expression on the cell surface. Could anyone please direct me to a publication (Or even unpublished observations) that described a depletion of pDCs when mice are injected with CD4-specific antibodies?
NO. The CD4 antibody shall deplete all the CD4+ T cells including the T regs and and T helpers, it will not effect the plasmacytoid DCs. CD11c antibody could be used to deplete the pDCs (if the subject is murine or in human peripheral blood anti-DLEC/CLEC4C/BDCA-2 could be used).

Firstly, is it your personal observation that CD4-antibody injection does not deplete pDCs? If so, do you have specific data you could possibly share?
Secondly, what is your opinion as to why it wouldn't?
The third one is actually just a comment. I am using PDCA-1 antibody to deplete pDCs instead of CD11c antibody for specificity.
Well I was concerned about the fact that its is the preDCs which are CD4 + that you are depleting but not the matured ones....as a citation found:
"Pre-pDCs express CD4 but lack T-cell receptor alpha (TCRα), TCRβ, TCRγ, TCRδ, or CD3 chains. Furthermore, they do not express B-cell lineage (CD19, CD21) or myeloid (CD13, CD14, CD33) markers. Pre-pDCs typically mature and produce large amounts of alpha/beta interferons (IFN-α/β) in response to viral and bacterial stimuliT cell subsets that would be targeted by the anti-Cd4s,
Also Murine pDCs differ from their human counterparts in expressing low to intermediate levels of CD11c, with lymphoid tissue pDCs also expressing CD4 and/or CD8α. Furthermore, murine pDCs from the spleen, but not blood, secrete interleukin-12 (IL-12) (p70) after stimulation with Staphylococcus aureus Cowan (SAC), in contrast to human pDCs."

So the anti-CD4 is working in your context.
For those who follow this question, our research group found that murine plasmacytoid dendritic cells in the spleen are depleted by repeated injections of anti-CD4 or anti-CD8alpha in a dose-dependent manner.
