I would recommend you to find the media that's specific for growing halophilic bacteria. "Handbook of Microbiology media by Ronald M.Atlas' would be a good place to start. In my experience, I found the book to be useful. Upon isolation, you may perform sequencing to confirm the identity of the type of bacteria. Good luck.

From your question it seems that you have the isolates and you want to identify these isolates. If yes then "go for 16S rRNA gene sequencing".

please carry out 16S rRNA gene sequence analysis. Sometimes, the normal 27f and 1492 may not click, if your isolate is bacteria, if archaea, sometimes 9f and 1492 may not click. if have not got success then use the primer set 530f and 1492r to amplify nearly 1 kb and determine its seq. followed by analysis at EzTaxon site. It will definitely provide you answer.
