I will prepare fibrin gel from this product (T1063/250 UN), and need to solve this product in 50 UI/mL concentration.
Please can anyone describe: how should it be calculated and diluted to 50 UI/mL, which solutions should be used for solving and diluting process, which conditions should be prepared before solving.
If UN stands for Unit NIH (NIH used to have their own measurement system separate from the WHO, at some point standards of each were prepared to make the NIH methodology of prep to yield 1 IU, same with the WHO)...it is equivalent to WHO Units (IU).
So, if that is the case, 250 UN = 250 IU. Dilute in 5 mL of water (can use 0.1% BSA as a stabilizer).
As an aside, the number that is actually important for activity is going to be a kahal unit (and nkal)

In this comparative study, from National Institute for Biological Standards and Control,1 NIH unit compares to 1.15 IU.I strongly recommend to dilute thrombin in a platic tube using 0.15 mol/l NaCl and 0.1% BSA. Thrombin is a rather sticky protein and adheres easily to surfaces. The BSA prevents adherence.
Lyophilized thrombin from the National Institute of Biological Standards and Controls (NIBSC) might be reconstituted with H2O, immediately followed by mixing with 5% human albumin of drug quality, immediately followed by 25% final conc. of glycerol. Thrombin from Sigma might be treated similarly.
