Our projects are to develop 'Halal' recommended media for Lactobacillus plantarum which doesn't contain pepton. We try to replace MRS with Skim. In order to make subculture media, we also think to use Skim Agar media. Can anyone help us know how to make skim agar media ? Best Regards.

Skim milk agar was found more types for any work it used .
in casein analysis was used 10 g skim milk powder in 100 ml water and add 2 gram agar , pH 7.2 when it used for proplytic bacteria count was the media preparation 100 ml nutrient agar with 10 % skim milk liquid (12 gram skim milk in 100 ml water) pH;7
I had prepared skim milk agar once. i added 10% skimmed milk powder to nutrient broth. you can remove the peptone from the broth and use only yeast extract
in practice you can sterilized the skim and other media (w/o pepton) separately, and then mix after sterilization.

I used to prepare the skim milk agar as Tony Stott (link above) describes and it worked well. Prepare the doble strenght "Nutrient Agar" (without peptone) from individual components.

I prepare 15 gram agar/L, autoclave it and adding 1% skim milk agar (pasteurized) after sterilization process

Separately autoclave the nutrient Agar then add sterilized skim milk in microwave to check proteolytic activity of bacteria.

In my view substitute peptone in nutrient agar with casein hydrolysate. It must work with the target bacterial specie.
