Does anyone knows the sonication buffer for paramecium/ protozoa , for two dimensional gel electrophoresis?

Hi!

I have to do 2 DE of paramecium total cell lysate. I am using

7 M urea, 2 mM thiourea, 65 mM DTT, 10mM Tris-cl, 4% CHAPS, 5mM PMSF, 1mM EDTA, for sonication, but not getting clear results on 1D SDS. Does anyone has optimized protocol for this.
