I am using Cyclin B1 as a marker for G2 phase in mouse embryonic tissues. However, when I use my anti Cyclin B1 rabbit monoclonal antibody, I only see signal in the cytoplasm of the cells. The protein is supposed to be localized in the nucleus. It also seems that all of the cells are stained and there is no clear expression pattern. I have attached a photo of the results. The area is the ventricular zone in the mouse embryo day E11.5.

Some more specifics:

The tissues were frozen.

They were post-fixed in 10% formalin for 8 minutes.

They were permeabilized for 10 minutes in 0.2% Triton in PBS.

I conducted antigen retrieval using Citrate Buffer pH 6.0 at 95-100 degrees for 15 minutes

I blocked with 2% BSA for one sample and 10% normal donkey serum for another. The results were the same

I got the primary from abcam and diluted it 1:500. The tissues were incubated overnight in 4 degrees in a humidifying chamber.

I used an alexa fluor 488 conjugated donkey anti-rabbit secondary and diluted it 1:500. The tissues were incubated for 1 hour at rt in the humidifying chamber.

I had a negative control that was secondary only. There was no signal in this control.
