I installed a superdex 75 pg myself, and the conductance baseline was flat at the beginning of the balance, and then the baseline was flat when the sample was eluted to 50%, and the last 50% jittered up and down, which was unstable.
To troubleshoot the issue and identify the specific cause of the uneven baseline, you can try the following steps:
Repack the Column: If the column packing is suspected to be the issue, you may consider repacking the column according to the manufacturer's instructions or seek assistance from the manufacturer's technical support.
Optimize Sample Preparation: Evaluate your sample preparation method to minimize interactions with the column material. This may involve adjusting buffer conditions, pH, or using additives to reduce sample adsorption.
Check Column Integrity: Inspect the column for any visible damage or irregularities. If you suspect column damage, contact the manufacturer for guidance on potential solutions.
Verify Instrument Setup: Ensure that your instrument is properly calibrated, and all components are functioning correctly. Check for any leaks or air bubbles in the system that may affect baseline stability.
I can also advise you to write to those support where you bought the device.
