Dear Gianluca,

The mathematical explanation for error-prone PCR and saturation mutagenesis was investigated by Patrick and Firth. In this respect Yuval Nov has added another viewpoint to the combinatorial complexity of such libraries and the probability if they can be targeted completely or not in the lab.

The group of Prof Reetz had several papers that look at this from the practical side, focusing on saturation mutagenesis.Investigating if the mathematical predicted completness is generated in a mutagenesis experiment.

The group of Schwaneberg has published a related case-study for error prone PCR.
