I know that few years ago a couple of papers were published about random mutagenesis of enzymes stating that, even using very high-throughput approaches, the number of clones that could be screened represented only a very small fraction of the all potential clones that could in principle be produced. The negative conclusion was that chances to find an interesting clone using completely random mutagenesis were very very low.
Now, I'm not able to find again such papers. Anyone have suggestions about?
