We've bought alpha-thrombin (from sigma T1063) for apta-sensensor study that have a concentration of 1099 NHI units/mg protein. I'd appreciate If someone help to convert this value to molarity.
All depends on the thrombin specific activity. If it is pure, specific activity is expected to be of 2,500 to 3,000 NIH/mg.
Based on 2,500 NIH/mg your preparation at 1,099 NIH/mg protein is about 40% pure (or active). It contains actually about 0.4 mg active thrombin, which has a MW of 36,700 Da (for the alpha form). This makes a concentration which is approximately of 10 µM.
