I am trying to grow WT Arabidopsis thaliana ecotype Col-0 in a growth chamber. The condition i use are 16hr day/8hr dark cycle at 22°C, 50-60% humidity and 10 Philips white fluorescent tubes for light. I transfer 14-days old seedlings from 1/2 MS media to moist autoclaved soil and acclimatize using plastic wraps for a week (poke holes for aeration). The soil that i used are plant-compost+ vermiculite. However, all of my plants died after acclimatization, although there's no fungus growth. Can anyone help me troubleshoot this problem? is it because of low light intensity? For light measurement simply use a photometer and optimize it accordingly (distance from source or number of lights). Acclimatization is a tricky job and several other parameters are considered as well i.e. Nutrients, water and gases level.

The term acclimated is used when the in vitro plants can survive the normal environmental conditions. How many days elapsed from the beginning of acclimation to the moment for which you considered the plants acclimated? Could be possible that plants aren't completely hardened?
You may use high humidity and supplement NPK nutrients. Initially 8h d and 16h n is good to check if it has effects
Dear all, thank you for the suggestions given. your help are very much appreciated. @ Papri Basak , you're right tht my soil was lacking enough aeration, so i changed the soil to peat moss compost+perlite and now my plants are growing happily. thank you!
