I am convinced that my enzyme is inhibited by atmospheric oxygen. One supporting evidence is that translation of this protein is only seen in hypoxic conditions. I want to support my hypothesis. any suggestions?
If you think the activity of your enzyme is inhibited my molecular oxygen, run the enzymatic assay in a glove box with controlled oxygen percentage. The evidence you state tells you that the gene transcription is inhibited by oxygen, but it does not necessarily imply that O2 will bind to the mature enzyme.

Do you want to see if the gene promoter is regulated by the oxygen concentration or if the enzyme is inactivated by O2?
in the second case, take a look in works done with hydrogenases / nitrogenases.
If available, you can perform the reaction in an anaerobic jar (wo O2) used for the culture of anaerobic micro organisms. Good luck with your research!
