Dear all,
I'm searching for a restriction enzyme that only cuts methylated bacterial DNA, just like DpnI? DpnI is not an option since it has too much cut sites in my plasmid.
I need such an enzyme since I want to select the ligation between a parental plasmid and a synthesized oligonucleotide. Just like in q5 mutagenesis protocol, but preferably with sticky ends.

The best option is call up the tech support of the company where you get your restriction enzymes and they would help you out

MspI is not the enzyme I'm searching for.
And I already tried NEB but their answer was not really what I expected. They suggested MspJI, FspEI and LpnI.
