The method in the paper that you have asked for is such that, you have to add 0.1N HCl to the Hemoglobin solution to acidify it to an extent it reaches pH 2.0, then add ice cold methylketone to it. shake it vigorously and keep on ice for 5 minutes and you will have aqueous fraction which has globin and ketone supernatant containing heme.
I have done this and it works properly. I just add equal volume of 0.1N HCl to hemoglobin solution and follow the same procedure as above.
Has anyone tried to remove the heme from separate subunits alpha or beta of hemoglobin? Just curious what was observed. I did heme removal on hemoglobin using the acetone method and it worked perfect.
