I'm trying to monitor the expression levels of a single component of a chromatin remodeling complex at work in the nucleolus of Arabidopsis. Any tricks out there for enriching this tiny organelle? Sucrose gradient?

Thanks for the suggestion. I am working with plant cells which have the added hurdle of a cell wall in addition to the cell membrane. I wonder if I can modify this protocol to include grinding after liquid nitrogen flash freezing.
