I need to cut sections of unfixed gonadal fat to stain for beta-gal. Fixation before embedding is not recommended when doing beta-gal staining. Unfortunately, in that way cutting a entire fat section is a really hard task. The tissue is so soft and seems to melt down when the blade goes through it in the cryostat. I tried to cut 20um sections at -30oC but still couldn't make a single good section. Any advice is more than welcome.
