We are trying to extract DNA from a bactiophage and are having issues with our DNA extraction kit (Promega Wizard DNA Cleanup System). We think the phage capsids are not lysing. Does anyone have any suggestions for enhancing phage capsid lysis?
you need to prepare strong lysis buffer
i dont think the one with the kit will do the job
There is a kit for SS-DNA purification from filamentous phage (M13 Spin minikit, Qiagen). The first buffer (PB) is able to lyse phage capsid. I dont know which kind of phage are you working with, but you could have a look.
Phage capsids are often very stable and don't lyse easily. You could try higher temperature with the lysis buffer or adding a phenol extraction step that should be very effective in lysing phage capsids.
Thank you all for your help. I will look into the Qiagen kit and possibly heating up our lysis buffer.
