We used this mixture of derivatization daily for quantitation of Testosterone.

The MSTFA products are stable unless you have some contribution of water or other solvent that has an "acid" proton like an alcohol. Which solvents do you use in between injections for clean the syringe? in which order?

We don´t evaporated the excess of derivatization agent because the excess assure us that all the Testosterone will be derivatize quantitative.

We have better reproducible result injecting two times a deritatize blank extracted from urine. It saturated the system.

Are you injecting spllit or splitless?

Which SIM ions are you looking for?

Thanks for your answer. I know that the reagent is really reactive with those components and I try to avoid them as much as possible. I am using Iso-octane as solvent for hormones and also to clean syringe between injections.

I am injecting splitless and focusing on Ions 432 (Testosterone), 434 (Dihydrotestosterone) and 416 (Estradiol).

reading again your question i have some doubts.

which solvent you use in between injections? because if it is a polar one it be consuming the MSTFA.

Another things is that sililated hormones there are not very stable in time. They hydrolyzed even in normal freezer. We prepare it daily.

Do you use deuterated internal standart?

I am using also iso-octane as cleaning solvent, so it shoudn't be a problem for the reagent.

I know the sililated hormones are not very stable, and it would be logical if concentration decreases in time, the problem is that after some time the concentration increases again.

Due to some problems with the instrument I couldn't do more experiments, but I will try as soon as possible.
