Thank you Ahed,

I have tried fixation with formalin, ethanol, mehtanol and acetone followed by staining but am not getting the desired results. Is permiabilisation step required? Al though it appears that acetone or ethanol fixation do not require permiabilisation it is not working for me.... any ideas
