Currently attempting to expand hIPSCs after thawing. Is there a basement coating for Geltrex that sustains feeder-free hIPSCs better? My current protocol (briefly) looks like this:

Well coating:

0.1% Gelatin incubation 37oC, 1 hour

1:100 Geltrex incubation 37oC, 1 hour

After the first thaw, all of my cells died, curious if it's my coating conditions, if gelatin should be replace with something more robust like polyornithine, or if Geltrex isn't appropriate?
