Hi Everyone, I used the one step EDC coupling method to attach the NH2 modified forward primers (with the C6-T8 spacer) to the carboxyl group on the Magnetic nanoparticles. I got the coupling efficiency around 60-70%. However, my primers detached from the MNPs during PCR reaction and I can’t detect the biotin labeled amplicon on the MNPs after magnetic separation. But I can detect the amplicon in the gel electrophoresis of supernatant (left over after magnetic separation of MNPs). I used the MES buffer (pH6.0) for the coupling reaction and prepared EDC freshly in MES buffer (concentration varied from 10 mg/ml to 100 mg/ml) but problem remained the same. Can anyone suggest how to stabilize the covalent bonding between MNPs and Primers?
