My aim is to measure microRNA using Luminex from archived samples, so basically formalin fixed paraffin embedded, and I don't know if I can extract good quality (and stable) RNA from this kind of samples for this kind of technique. I've checked in literature, and RNA was successfully extracted from FFPE samples, but the most of the paper measure microRNA using other assays...
The Luminex technology is fairly tolerant to alternative samples but has an inherent variablity that will cause you to have to exclude a lot of your data anyway. That being said I know that fixed tissue can not be used for the protien based assays but my guess is more that it's due to changes in protien structure from fixation than the actual formalin. My suggesting for you is to run in triplicate if possible as values with a %CV>30 should be removed from the data set; and this will happen a lot. Millipore kits tend to be more flexible than the BioRad ones. Best of luck!
