Our lab bought the KingFisher Duo from Thermo Fisher for DNA extraction from saliva samples (human). We are using the MagMax Total Nucleic Acid Isolation kit (Thermo Fisher). I have been using it for some time now with really bad results.

Almost always, some of the samples ends up with beads in the final elution, sometimes so many that I can't clean it manually and have to discard it. I have tried everything but the only thing that seems to be related to this problem is the saliva consistency.

In addition, I am getting low DNA concentration in general, usually less than 50 ng/ul (Quibit cuantification). I use 400 ul of saliva (following MagMax protocol).

Has anyone had similar problems? Any insight and comments will be much appreciated!
