1- you can omitted macrophages by culturing PBMC in culture plate with SFM (serum free media ) for 30 min , after this time macrophages attached to plate .

As above, you can selectively remove monocytes/macrophages by incubation PBMC's in serum-free media for one hour. Monocytes will adhere to the plate allowing you to remove the remaining cells for co-culture.

If you decide to remove Macrophages, above mentioned technique will work fine. Question is, do you really want to remove Macrophage from PBMC culture ?

I think, your cell line morphological structure should have significant difference with PBMC macrophages. Eitherway, if your cell line is suspension cells, followed by few passage , you will get macrophage free cells suspension. On the other hand , if your cell line is adherent one, then you trypsine treatment duration for cell line & macrophage detachment should be different , which certainly will lead to macrophage free cell suspension for your experiment.

plastic adherence to deplete monocytes does not guarantee that you actually deplete all monocytes (most adherent cells will be monocytes, though). If these cells are of concern, you should use different approaches (e.g., beads)
