Can someone kindly provide me the detailed procedure to carry out desulfation and methylation analysis for fucoidans? There are many publications, but neither of them describes how to do it in detail (I was unable to find any standardized method). If someone knows of a pre-optimized method, it will be very much helpful and greatly appreciated.

The simplest method is by infra-red spectroscopy if not by HPLC after hydrolysis
Dr. Hatem Majdoub, Thank you for your answer. Yes of course FTIR is the most reliable method to analyze polymers such as fucoidan for their functional groups, connectivity of those groups etc.. Methylation analysis or the linkage analysis on the other hand is a method that is used to predict the connectivity pattern of monosaccharides (sequence of nearby monosaccharides). It provide details that we cannot get from FTIR. And using these details we can construct the rough structure of the polysaccharide backbone.
