I would like to do QPCR for metastasis for a cancer model expressing the SV40 Large T antigen.
Have you made primers that work for QPCR? I tried 3 primer sets from the literature that did not work and showed signal in negative control samples.
If yes please share these with me or refer me to your publication. Any help is appreciated.
I think the best option is design your own primers. Do you know which SV40 vector was used to establish the model you are working on? Most vectors used for cell immortalization have optimizations (deletions, mutations) or even truncated sequences of the SV40 large T antigen, making it troublesome to retrieve working primer sequences from the literature.
After my failed attempts using literature primers, I think you're right. Thanks for your reply!
