i culture Rosa shoot tip in MS medium at beginning it started very well but after 2 weeks it tend yellowish and finally necrotic. paper report that increase Iron to 20 percent and calcium 50 percent solve the problem i increase it but there is no good result. Is there anybody can help?

I'm agree with iron increase but, not the form that is used in MS medium, per my opinion, use FeEDDH instead of FeSO4 equal with Iron concentration in MS medium; wish to be of help!

Thanks. some paper reveal that its cholorosis is because of ethylene production in in vitro medium. can i use agno3 in tissue culture medium?
Not clear to me why you focus on iron in the medium. Can be another component. I would not expect the medium itself to release ethylene. Maybe the cultivar you use is more specific and then maybe you try another basic medium - e.g. WPM or another. Have you played with the plant growth regulators - types, concentrations, combinations..? Can be they or various other factors. It is difficult to advice you since we do not know what is your aim and what else you have tried.

Why you want to use a shoot tip? Are you trying to get virus free plants? Size of the explant too matters. Usually for micropropagation axillary buds are used. If the explant is producing phenolics, you can use phytagel/gelrite and/or maltose instead of sucrose. What hormones you used so that shoot tip grows faster and does not produce phenolics. You can use a micropore tape to seal the Petriplate so aeration will not be a problem.
thanks. I used the axillary bud and for establishment stage i use the BA 1mg/l and IBA 0/1 mg/l. but after around 2-3 weeks it start chlorosis and necrosis. agar-agar was used for gelling agent.
For Rosa micropropagation you should use FeNaEDDTA instead of FeNaEDTA for cultured medium or you can change MS vitamins by Gamborg vitamins (vitamins B5) for medium cultured Rosa. The development of shoot tips also depend on which genotype you use for in vitro culture. Some genotypes are very recalcitrant in vitro culture condition.
