Hi, I'm having a problem with the loading control of my WB where I'm comparing different cell lines. In the picture I have beta-actin on the top and GAPDH on the bottom and I'm comparing Jurkat, HeLa, THP1 and HL60, by this order two samples of each (one control, other with an added compound) and I need to compare a protein between the cell lines. The problem is that loading controls are not even, but it doesn't seem to be a loading issue. Jurkat and HeLa are fine, but then THP1 shows too much beta-actin and few GAPDH and HL60 it's the second pair of samples that I test and I always have very low levels or none visible beta-actin!!

I've read that different cell lines may have different levels of beta-actin, however we checked with other people that work with these cell lines and there shouldn't be any difference, much less such a big one! I'm also having this problem with cell line Molt4 (first lane on 2nd figure) where I have a lot of GAPDH but none beta-actin and U937 (3rd lane). Ponceau staining showed no significant differences between lanes.

Did anyone had a similar problem? Anyone knows what can be causing this? Thanks in advance!
