xylan from oat spelt can be procured from HIMEDIA Laboratories (India) and it gives good results during screening for xylanase producing microorganisms.....any further query feel free to ask....

For screening purpose, I recover crude xylan from breakfast oat meal.

I prepare the medium as per method given below.

1. Suspend 25 gram oat meal ( any brand from grossary store) in 200 ml distilled water at room temperature and mix it well by stirring for 15 min using magnetic stirrer.

2. Strain the contents fusing tea strainer.Store the passthrough on ice for 1 hr.

3. Transfer the retentate in the previous container add, about 100 ml water to the retentate and stirr agin for 15 min , repeat 2.

4. Pool both passthroughs and transfer the contents in a 500 ml centrifuge bottle store in the pass through in refrigerator overnight.

5 Next morning gently transfer the content into a fresh centrifuge tube without disturbing the sediment at the bottom. (You may sacrifice some content to avoid the sediment)

Spin 10,000g for 30 min. Transfer the supernatant into a autoclaving flask.

You can use this solution as a xylan solution. Add nutrients and buffers etc and make your medium which will have xylan as the major carbohydrate. This medium gives good resuts with Congo red assay also.

Well, xylan is, next to cellulose the most important biopolymer. Both, cellulose and xylan are polysacchrides that are found in plant cell walls to give them mechanic support, meaning stiffening them. Oat is a plant cereal. And xylan from oat spelt just means that this specific xylan was extracted from oat spelt.

we do have xylan from oat spelts in our lab. it was bought from sigma aldrich company. You can try!!

Sigma-Aldrich provide xylan from beechwood but they discontinue the supply of xylan from oat spelt. What is the difference between xylan from oat spelt and beechwood? Can anyone help me out? It is said that xylan from oat spelt is insoluble in water. But not sure about it.

Hey Dan, the main difference is, that xylans from wood are usually different xylans than those from grasses or annual plants, which are mainly arabinoxylans. We found these arabinoxylans work best for our screening purposes...

I worked with several different xylans before, and none of them was completely soluble in water. They do solve to a certain degree though, but there's no difference between hardwood, softwood or arabinoxylans. At least not for me.
