Does anybody know a better minimum promoter with almost no detectable leak that I can use in the GAL4-VP16/UAS system? I'm doing transient expression in tobacco and the 35S minimum promoter has a relative high basal expression.
You didn't give any background for what you are trying to do, but I think you might want a promoter that is not always on. In my work, I consider the 35S minimal promoter to be a constitutive promoter, meaning outside of gametophytic cells, it wouldn't be considered leaky, just always promoting transcription. Thank you for your suggestion. I am developing a system using GAl4/VP16 and UAS. I have shortened the 35S minimal promoter and it still promoted transcription. The assay I am working on is very sensitive even to a very low transcription driven by the 35S minimal promoter.
