As we know degranulation assay of NK cells is to check the NK cell functionality, so is there anyone doing NK degranulation assay after incubating the whole blood with antigens for 24hrs, then in the next day isolating the PBMCs and co-culture with K562 cell line?

I would prefer to isolate NK cells on the same day, as they are very fragile and present in a relatively small numbers compared to other cells, therefore separating them from the whole blood as soon as possible would give you higher viability. After the isolation, I prefer to subculture them for 2-3 passages before doing any assays, just to ensure they are in a good condition.
