Does anibody see monocyte killing when human pbmc are activated by soluble okt3?

I'm trying to study the impact of t cells on monocyte activation and polarization using human pbmc in vitro. As far as I use monocyte stimuli (e.g.TLR agonsists) it's fine, but when i trigger T Cells by soluble okt3, most monocytes die. Could this be an in vitro artifact? Does anybody observe the same phenomenon? Thanks for any feedback! Licia

Hi,

OKT3 is an IgG antibody and Monocytes express receptors for the Fc regions of gamma immunoglobulins. Therefore, one possible reason to explain the death of monocytes upon triggering of T cells with OKT3 Ab in your experiment is the crosslinkage of inhibitory Fcgamma receptors on monocytes. The binding of OKT3 to T cells forms immune complexes and the Fc regions in these immune complexes could crosslink inhibitory Fc receptors on monocytes leading to apoptosis of monocytes.
