Since Activated Charcoal is an inert solid adsorbent material used in plant tissue culture to improve cell growth and development as well as adsorption of inhibitory substances in the culture medium. It is said that AC alters medium pH to an optimum level for morphogenesis How the addition of inert AC will change the pH of the nutrient medium?.

The addition of activated charcoal could cause decreasing pH of the nutrient medium. Of course, it depends on AC concentration. As a part of a research, I recorded the pH of the MS medium containing different concentrations of AC after autoclaving. The results showed that the addition of AC in concentrations up to 500 mg/l could slightly decrease the medium pH (0.05-0.1 units), and the addition of AC more than 1 g/l up to 5 g/l could have a significant effect on reducing the pH of the nutrient medium (0.2-0.4 units).

@ Manokari, my observation also the same as Dr. Ruhollah, activated charcoal decreasing the pH in the nutrient media but definitely it depends on the type of activated carbon and the chemical composition of the media.

It seems the effect of activated charcoal on the medium pH is depends on the process and method of production of activated charcoal, such as the raw materials (plant species), the chemical agent used for the activation, etc.

Activated charcoal is normally added in cultures to absorb phenolics released by explants. The pH is already set before autoclaving the media so not much should be expected to change.

Despite the decrease in PH of the culture medium, these changes are not significant, so do not worry.
Ac improve root growth within two weeks . Along with Gamborgs and Activated Charcoal is improves the root length too. 5.8 pH is the best pH value for the in vitro plants.
