Hello, I am a postgraduate student in China. Recently I detected the viability of AML12 cells treated with APAP and its combination with protective agents. Unfortunately, the viability results (from cck8 test) were not reliable because the media became brown in APAP-treated group.

  • I understand it is more appropriate to use primary hepatocytes to test the toxicity of APAP rather than AML12 cells. But AML12 cells are what I can obtain currently. I am thinking about the reason of color change in media treated with APAP. Is APAP oxidated into another intermediate product with color?
  • Do you have other solutions to detect the viability of AML12 cells on this occasion? Could I detect the LDH from the supernatants of media treated by APAP?
