We are performing any soft agar colony assay. We use crystal violate stain. But now we would like to destain our plate and then observe the single cell colony of MCF-7 cell line. Can you please give me the destaining protocol for that. We are not able to get good results by using 1% glacial acetic acid+4% Methanol+5 % distill water.

Usually I destain it with just dH2O three times, for 30min with agitation and it works just fine. If the staning is too intense you may have to consider reducing the incubation time with crystal violet, or its concentration.

Pure Methanol streight into culture plate works as a very good destaining agent for crystal violet. You can opt for the same.
