Primordial germ cells show several features of motile cells. However some authors suggested that the migration due to morphogenetic movements and local cell divisions. Do these cells migrate actively or passively?I would like to share ideas about this subject.
Try adding 5micromolar Amlodipine to the flask. If you see a reduced motility the motility is actin dependent and is active.

Good idea, thanks. I already read about the in-vitro motility of PGCs ( confocal microscopic studies), but I still wonder about the active migration, could cells really migrate, do it needs another factors I mean fluid (inter-cellular), secretions or special ground substance (matrix)???.
There is plenty of research showing that primordial germ cells migrate actively within the tissue. In zebrafish, it was shown that PGC migrate against tissue morphogenetic movements ruling out passive migration. Germ cells are guided by the chemokine signaling and eliminating this signaling caused cells migrating non-directionally. Also, blocking actin polimerisation, either through calcium signaling as previous commentator already suggested, or by other means, and other cytoskeleton regulators render cells immotile. In that case they were indeed dragged along the morphogenetic movements in developing embryo. In culture, however, it is very difficult to replicate the motile behaviour of these cells since they rely on the surrounding tissue for adhesion and signaling. Hope that helps.
In chicken PGCs migrate through blood. But in mouse (as mammalian models) its migratory road is full of mesenchymal cells, cells every where, the migration occur rapidly without the presence of surrounding fluids these slightly disturbed me. I have been understand the role of cytoskeleton and the tropic and other regulator factors. however, is there a role for the matrix or the interstitial fluid in this migration??? body fluids and or the matrix usually aids wondering cells.Is there a suggested methodology to evaluate migratory PGCS in-vivo ??.
