I'm working with fixed brain tissues in 4% paraformaldehyde, using sucrose and antifreeze as post cryoprotectors. I only work with fresh solutions, made in the day of use, so I'd like to know if I have to put this solutions in ice while I'm working with them, as I take a while to substitute one solution for another (~ 40 minutes).

You don't need to put sucrose solutions (such as sucrose 15% or 30%) on ice while you are working with them...However, you can consider the option in case you need to work with them for a long time.
Normally, for 15 - 20 samples, I don't take more than 10 min to substitute paraformaldehyde with sucrose 15% or sucrose 15% with sucrose 30%...In this conditions, I don't need to put my solutions on ice.
