Do I need glycerine to weigh down the samples of DNA/ Protein I will be running in a gel? Does it matter if it is vegetable or animal glycerine?

You can use sucrose as well. You need some sort of reagent to increase density of samples so that they can stay in the gel wells. Now for animal or vegetable glycerol, honestly I never heard that it matters where it comes from.
