From what I know, the virus acquisition and transmission rate is reduced greatly when aphids stay on non-persistent virus infected plants. Does that mean that when the aphids are reared on such plants or colonize them they do not acquire the virus very well?
The answer is yes. The virus will be in the stylets.
The answer is no. The non-persistent virus will not be in the salivary glands. It may be in the gut, but just passing through.
The answer is maybe: It may depend on the virus in question. The old paradigm is that non-persistent virus is transmitted as a contaminant. The aphid feeds on an infected plant and a few virus particles happen to adhere to the inside or outside of the stylets and when the aphid moves to a new host these contaminants get transferred to that host. However, some non-persistent viruses cannot be transmitted when specific parts of the virus genome are missing or changed, and parts of the insect genome also influence transmission rates. These observations are generally inconsistent with the old paradigm. At least in some cases there is clear evidence that virus adheres to very specific areas of the insect stylet. Your choice if you consider this "in their body" or not.
