Hello, we are in beginning NGS projects with rhizosphere soil using Qiagen's powersoil extraction kit. In past labs, I have performed DNA extraction in several locations: chemical fume hoods, laminar flow hoods, open benches in laboratories, as well as dead air boxes (DAB) (all areas were sterilized with bleach/h2o/EtOH prior to beginning the extraction).

Due to the several different lab places outlined above used previously for DNA extraction, I am wondering if there is a general consensus on best location to preform extraction. We use PCR hood / DAB for qPCR and PCR plate-loading, and am open to this space for DNA extraction but ultimately prefer to keep separate. Additionally, both negative and positive controls will be included no matter the location.
