Hello, we are in beginning NGS projects with rhizosphere soil using Qiagen's powersoil extraction kit. In past labs, I have performed DNA extraction in several locations: chemical fume hoods, laminar flow hoods, open benches in laboratories, as well as dead air boxes (DAB) (all areas were sterilized with bleach/h2o/EtOH prior to beginning the extraction).
Due to the several different lab places outlined above used previously for DNA extraction, I am wondering if there is a general consensus on best location to preform extraction. We use PCR hood / DAB for qPCR and PCR plate-loading, and am open to this space for DNA extraction but ultimately prefer to keep separate. Additionally, both negative and positive controls will be included no matter the location.

Giorgia Pertile Thank you for recommending DNA-extraction service labs. However, we plan on doing in-house extractions and I am inquiring about which lab space to use to complete the DNA-extraction workflow. (e.g. laminar air flow hood, chemical fume hood, open air bench, dead air box/PCR hood, etc.)
If you have enough rooms, use 3 rooms: one for DNA extraction in a PCR hood, another to load that DNA and a third one for PCR ampification. If you have available and want to prevent initial contamination use the laminar flow hood. It all depends on your objectives.
