DNA barcoding on free-living marine nematodes is very difficult. At the same time, some researchers have done this group using 16s rDNA / 28s rDNA. But to my knowledge, no one has done in COI? Does anyone know the procedure of DNA barcoding on free-living marine nematodes using COI gene?

CO1 based DNA barcoding protocol is same except that what primers are successful.....my suggestion is go to BOLDSystems and find out the protocols as well as the primers under nematodes
yes COI gene has been used previously by many researcher. Folmer region is not very good region for Nematode barcoding but I3- M11 region is good for the same.
