Hi all,

Could you please explain what is the difference between the replication origin pBR322 and ColE1 ?

If you have look on that vector databases: http://www.embl.de/pepcore/pepcore_services/strains_vectors/vectors/bacterial_expression_vectors/popup_bacterial_expression_vectors/index.html or https://www.embl.de/pepcore/pepcore_services/cloning/choice_vector/ecoli/vectorfeatures, we can see that, for instance, pET-3a is indicated to have a pBR322 origin. However, after SerialCloner analysis, this is the ColE1 origin that is detected.

Is it the same origin? If yes, why some vectors are designated to have ColE1 OR pBR322 in this database ?

Same question with pUC origin. When I look at the pUC57 vectors (for instance), I found a ColE1 origin….

Again, is it possible that several replication origin are on the same vector ? For instance, on pET-28b+, I found ColEI, M13 and F1 ?

Thank you very much for your expertise.
