I'm using the very same stock of S.aureus but, in the time course of my infection experiments in human MDMs, the CFU absolute number in the control cells is contunuosly decreasing.
Of course the human macrophages are killing S.aureus, the problem is the extremely low CFU count I'm getting since a certain date in the untreated cells as compared with the activated ones. I know that have to consider the donors variability (they are primary monocyte-derived-macrophaes from healthy donors whole blood) but I have the suspect the the new S.aureus stock is not growing as the previous one. Any idea?
