I started the analysis of SOD (superoxide dismutase) activity in human urine using ELISA. The kit manufacturer only provides reference values for serum so I'm not sure if the values I get are ok. It is also hard to find something in the literature. Could anybody help?

What you can do is order standards and dissolve them in a buffer solution to get a calibration curve. With the function you get out of that you can calculate concentrations. Whether or not urine interferes with your kit is not something I would be able to tell you however makeing a calibration line in phosphate buffer is a valid way of approaching this assay.

I am not aware of any measurements (values) for SOD activity in Urine! Do you know if it is MnSOD, CuZnSOD, or Extra cellular CuZnSOD? For which SOD does the ELISA kit you are using specify! I suggest that you characterize the SOD in the urine (using biochemical isolation and determining which isoform are you dealing with) then design a custom made ELISA for the assay.
From the publications recommended by Peter Kroneck, I see that CuZnSOD (SOD-1) is the major SOD in Urine. Therefore, I suggest using Westerblots as in Paper #2.

Thanks a lot for your response. @Hosni There are commercial kits to measure Cu/Zn SOD http://www.enzolifesciences.com/ALX-850-033/cu-zn-superoxide-dismutase-elisa-kit/. The problem is the most reference data in humans is for serum. Since I'm doing this for the first time I wanted to make sure that what I get is right!

I am trying to learn a lot more about SOD, particularly the role of bacterial SOD in humans (during sepsis, bacteremia, cellulitis, etc., for example) and whether that can be measured too. I would be grateful if everyone here could also keep an eye out for topics covering this subject too.
