I want to measure the uptake of Flagellin after treatment to the cells.
Please could anyone having experience for this experiment give me some suggestion. I tried to detect by Intracellular staining (FACS) but it seems not much sensitive and stable so I could not get the consistent results.

In my experiment, first, I added flagellin to cell culture and then measure flagellin uptake by Primary antibody against Flagellin follwed by secondary antibody conjugated PE.
So I just wonder, what is difference between PE conjugation and FITC conjugation and how to decide which one is more suitable for my experiment.
If you have some experience, could you give me some advice.
