I have Balb C mice that infected with Schistosoma mansoni, I am trying to detect IL4 cytokine using supernatants from SEA and CON a stimulated spleens using cytokine elisa, but there is nothing showing up, I have a recombinant IL4 as the standard and control. Any ideas what I might be doing wrong cause I can detect IFN-g with no problem. the Antibody pairs are 11B11 clone and BVD6-24G2.
This might be a kinetic issue i.e. depends on the time you are analyzing your cytokine response. Try detection of the cytokines from 0 to 10 hrs after activation.
For how long did you stimulated your splenocytes? I can be that 72h is not the optimal time point, you can try 48h stimulation.
But since you got IFNg production, it could be that you dealing with a type 1 response and you will not measure any IL-4 at all.
IL-4 with helminthes is quickly degraded so no detection or detection with low volume.
