First, I should make a slurry of C18 silica in methanol. Then I pack the column and then do several washes with methanol. For adaptation of the silica by the first mobile phase, I'll use 10% methanol in water and then load the sample, which is soluble in this mobile phase or adsorbed on C18 silica (dry loading). Then I'll start the elution by 10% methanol, 20%, 30% until 100% methanol is reached.
