can anyone share a method about how to destain the giemsa staining?
I fixed cells by carnoy's fixative and dropped on the slide to produce the metaphase spread and stain it by Giemsa. Now I need to destain Gimesa and stain the centromer of chromosomes.
I have tried to put the slides 3 mins in each 95% ethanol, 70% ethanol, aqua dest. But the only a few cells are destained. most of them are still purple.
I also put the slides in 100% methanol for 3mins. it looks better but I am not sure if it is okay?
Methanol is best to detain the Giemsa staining. use it for few minute to destining.

Try to destain with methanol(absolute or 95% solution). It works for destaining gimsea done on histological sections or blood smear.

I can recommend fixative solution (methanol and acetic acid 3:1) for 2-3 min at RT. Easy and fast.
