I am investigating on isolation and identification of cry genes in Bacillus thuringiensis.

After isolation Bacteria according to the related papers, gram staining was conducted. Also, commassie blue used and the bacteria were checked for crystals. Then, a bioassay was performed on Ephestia kuehniella and the results showed that several bacteria caused mortality on the floar moth even more than Bt subsp thuringiensis. But, for PCR by primers, no gene was detected. What is the reason?
