Hello All,

I am currently working with ExpiCHO cells to express proteins which can go up to a cell density of upto 11 x 10e6 cells in a serum-free environment.

Lately, the cells grow slower, and when they reach a density of >6 x 10e6 cells they die.

I revived frozen vials and they also behave the same.

Temperature, CO2, and media is not a problem. If there was contamination I would be able to see (not checked for Mycoplasma - Could it be that>?????)

The challenge is the viability will be >90%. and after 36 hours it is 1% and all cells died with no E.coli or fungal seen under the microscope.

Has anyone experienced sudden crashing of culture? What can I suspect and try to pin point the problem.
